Development and validation of a full scan method to determine blood alcohol concentration by gas chromatography with detection of simple quadrupole mass spectrophotometry

Abstract

Blood alcohol concentration in one of the most frequently performed chemical tests in forensic laboratories, so it is essential to have accurate, precise and fit-for-purpose analytical methods. The objective of this study was developed and validate a method to determinate blood alcohol levels by gas chromatography/mass spectrometry. The method was developed with headspace injection, using propan-1-ol as internal standard, the gas chromatograph was set at 40 ºC, the mass spectrometer data acquisition was full scan (28 - 65 uma), the analytes were identified by comparison of the mass spectra with the mass spectrometer libraries, ethanol was quantified considering the ionic fragment 31. The developed method was selective (it efficiently separated ethanol from methanol, ethanal, propanone, propan-1-ol and propan-2-ol with a resolution greater than 3), sensitive (the limits of detection and quantification were 0.00213 g/L and 0.00710 g/L respectively), linear (R2 = 0.9994, in the working range from 0.1 g/L to 4.0 g/L), precise (the coefficients of variation were less than 1.36 % and 1.97 % in the repeatability and intermediate precision respectively), accurate (the recovery obtained was from 98.4 % and 101.7 %) and robust (statistically the same results were obtained when by making minimal variations in the method). It is concluded that the method developed and validated for the determination of alcohol levels by gas chromatography/mass spectrometry is selective, sensitive, linear, precise, accurate and robust.
