Effect of freezing method and incubation period on post-thawing quality of equine semen

Abstract

Semen cryopreservation produces a high degree of alterations in the sperm that generate a consequent loss of fertility and a lower pregnancy rate when compared to fresh semen. Due to this, the present study aimed to evaluate the effect of the freezing method and the incubation period on the quality of equine semen after thawing. For this, semen was collected from three horses using an artificial vagina. The semen was subjected to two freezing methods: nitrogen vapours and programmable freezer. Once thawed, the semen was kept at 37 °C in a water bath and incubator, and semen quality evaluations were carried out at 0, 1 and 2.5 h. It was determined that the use of nitrogen vapours turns out to be the most suitable for cryopreserving equine semen and that maintaining the thawed semen in an incubator turned out to be much more suitable compared to incubation in a water bath.
